High-Throughput Intact Covalent Binding: Analysis Automation Workflow for Scaled Multiplexed Screening with Stringent Control of Quality

Recent interest in covalent drug discovery has highlighted the need for robust, high-throughput analysis methods, only achievable with automation. This method maximizes inherent benefits of intact MS analysis. It enables deplexing of compound mixtures, increasing throughput whilst decreasing cost. It also identifies non-specific binders much earlier than traditional screening.
Here, we show a fully automated data analysis workflow and quality control of multiplexed intact covalent binding MS screening. This enables such screens at high-throughput scale typical in industrial discovery.
